SamNor:  A. actinomycetemcomitans was detected in 52.3% of the patients
          by the latter technique while the AffirmTM DP test failed to detect
         the bacte in any of the samples. Overall, the results of the present
         study confirm the importance of the screening method and indicate tha
         t the prevanlences of the investigated putative periodontal pathogens
          and beneficial species in the healthy and diseased adult Cameronians
          were similar to those reported for adults in the West and in some de
         veloping countries.
SamEng:  Our study is the 1st report on subgingival microbiota in adult Camero
         nians. The aim was to investigate, using the checkerboard DNA-DNA hyb
         ridization technique, the prevalence of 18 oral species in subgingiva
         l plaque samples obtained from sex- and age-matched Cameronian adults
          with and without periodontal destruction. We also compared cultivati
         on and the AffirmTM DP test with the checkerboard technique in their
         capability to detect some selected species among the 18. 21 adult per
         iodontitis patients and 21 periodontally healthy subjects were examin
         ed and the reults were compared statistically. Each periodontitis pat
         ient had at least 4 pockets of 6 mm depth, while the healthy subjec
         ts had no sites more than 3 mm deep. Results of the checkerboard anal
         ysis showed that significantly (p<0.05) more periodontitis patients t
         ested positive for most of the 18 bacterial species. The Gram-positiv
         e species Actinomyces naeslundii, Streptococcus mitis a
         nd Streptococcus sanguis, known as microbiota of healthy sites
         , were detected significantly more frequently in the healthy group. C
         ultivation demonstrated P. gingivalis, B. forsythus, A. actinomyce
         temcomitans, P. intermedia and F. nucleatum in significant
         ly lower %s of patients as compared to the checkerboard technique. Fu
         rthermore, the AffirmTM DP test detected P. gingivalis and B. f
         orsythus is significantly fewer patients than did the checkerboar
         d technique.

SamNor:  The resulting data for the 2 enzymes were combined and subjected to c
         luster analysis using an average unweighted linkage sort. In some ins
         tances, isolates that appeared to be of identical ribotype using one
         endonuclease gave different ribotypes using the other. Strains of 
         P. gingivalis within a subject were usually identical, except for
          3 patients who harbored 2 different ribotypes/individual. All subseq
         uent analyses employed a single ribotype strain for each subject. A t
         otal of 32 ribotypes were observed for isolates from distant countrie
         s. A total of 11.5% of the patients had isolates exhibiting the same
         ribotype: ribotype 7a. Identical ribotypes of P. gingivalis ca
         n be recovered from subgingival plaque samples of patients in differe
         nt countries.
SamEng:  Porphyromonas gingivalis has been isolated from periodontitis
         lesions in subjects from many geographical locations. The purpose of
         this investigation was to determine whether similar ribotypes of P. g
         ingivalis could be detected among strains isolated in different count
         ries. A total of 198 isolates of P. gingivalis were obtained f
         rom 52 periodontitis patients in Boston (130 isolates), Bergen, Norwa
         y (17 isolates), Khartoum, Sudan (26 isolates), and Bucharest, Romani
         a (25 isolates). DNA was isolated from each strain, cut separately by
          the restriction endonucleases KpnI and PstI. The resulting preparati
         ons were subjected to electophoresis in a 0.8% agarose gel using a Tr
         is-acetate EDTA buffer. Uncut lambda and a 1000-bp fragment of 16S rR
         NA were included as internal standards in each lane. In addition, a H
         indIII digest of lambda was present in a separate lane in each run. T
         he DNA fragments were transferred to a nylon membrane by downward cap
         illary transfer. 16S rRNA bankds were detected using a 1000-kb digoxi
         genin-labelled probe generated by a polymerase chain reaction. At the
          same time, a digoxigenin-labelled probe to lambda was employed to de
         tect the internal and molecular weight standards. The bands were dete
         cted using antibody to digoxigenin conjugated to alkaline phosphatase
          and chemiluminescence. The positions of the bands relative to the in
         ternal standards were determined and normalized to correct for run-to
         -run variations, and the molecular weight of each band was determined
          by comparison with standards within each gel.

Tittel:  A study about some Prevotella isolates from periodontal abscesses.

SamEng:  The aims were to characterize biochemically and to speciate Prevotell
         a isolates from periodontal abscesses. Gram-stained smears and cultur
         es were performed with pus samples collected by needle aspiration fro
         m periodontal abscesses of 25 Romanian patients. Cultivation was done
          on Columbia blood agar (aerobic incubation at 37 degr. C for 24-48h)
         , and on Fastidios Anaerobe Agar and kanamycin-vancomycin laked blood
          agar (anaerobic incubation at 37 degr. C for 2-5 days). The isolates
          were identified on the basis of Gram staining, cultural characterist
         ics and standard biochemical methods. The Gram negative anaerobic bac
         illi were biochemically characterized and identified at species level
          using the Rapid ID 32 A system (Bio Merieux, France). Prevotella rec
         overed from 15 samples. Speciation showed: P. melaninogenica (8 isola
         tes), P. dentioda (3 isolates), P. oralis (2 isolates), P. loescheii
         (1 isolate) and P. bivia (1 isolate). All Prevotella isolates tested
         similarly in 20 tests of the Rapid ID 32 A system. The P. melaninogen
         ica strains reacted differently only in 2 sugar fermentation tests. C
         onclusions: Prevotella is often involved in periodontal abscesses (>
         50% of the cases), P. melaninogenica predominating among the identifi
         ed species. The biochemical profile of the 15 Prevotella isolates var
         ied less in the Rapid ID 32 A system.

SamEng:  Mucosal immunization can induce immune responses different from those
          induced by systemic immunization. In this study, murine immune respo
         nses were analysed after immuniation through gastric and subcutaneous
          routes with Mycobacterium bovis BCG. The number of circulating cells
          secreting antigenspecific immunoglobulins (Ig), and the number of ga
         mma interferon (IFN-) secreting cells in spleen-cell cultures after
          in vitro stimulation with myucobacterial antigens were analys
         ed at the single cell level by the ELISPOT method. Levels of antigen-
         specific antibodies in sera were determined by ELISA. In the s
         ubcutanously immunized mice the authors found approximately 100 times
          more splenic cells secreting antigen-s pecific IgF than in gastrical
         ly immunized mice or control mice. Their levels of antigen-specific I
         gG in sera were 66-6700 times higher than in sera from gastrically im
         munized mice orcontrol mice. In contrast, the numbers of IFN- produ
         cing cells in spleen-cell cultures after in vitro activation w
         ith BCG were equally high in the immunized groups of mice, and for bo
         th groups higher than in non-immunized controls. Furthermore, IFN-
         producing cells could be demonstrated in gastrically immunized animal
         s even without in vitro activ ation. The results demonstrate t
         hat gastric immunization with the BCG vaccine can induce a systemic T
          cell-mediated immune response against mycobacterial antigens.
